Evidence for yeast artificial synthesis in SARS-CoV-2 and SARS-CoV-1 genomic sequences

Background: Knowledge about the origin of SARS-CoV-2 is necessary for both a biological and epidemiological understanding COVID-19 pandemic. Evidence suggests that proximal evolutionary ancestor belongs to bat coronavirus family. However, as further evidence direct zoonosis remains limited, alternative modes biogenesis should be considered. Results: Here we show genomes from SARS-CoV-1 are differentially enriched with short chromosomal sequences yeast S. cerevisiae at focal positions known critical host cell invasion, virus replication, immune response. For SARS-CoV-1, identify two sites: one start RNA dependent polymerase gene, other spike protein’s receptor binding domain; SARS-CoV-2, viral replicase domain, toward end gene past its domain junction. At this junction, detect highly specific stretch DNA covering furin cleavage site insert PRRA, which has not been seen in lineage b betacoronaviruses. As natural family, propose passage model constructs cells based on co-transformation plasmids carrying selectable genetic markers followed by intra-chromosomal homologous recombination through conversion. Highly differential sequence homology data across chromosomes congruent harboring auxotrophic support model. Conclusions: These results provide among SARS-like coronaviruses only contain information points synthetic genetically modified cells. Our specifically allow identification cerevisiae as potential donor SARS-CoV-2.
